Neuropathic pain is one of the most intractable diseases. The lack effective therapy measures remains a critical problem due to poor understanding cause neuropathic pain. aim this study was investigate effect dexmedetomidine (Dex) in trigeminal and underlying molecular mechanism order identify possible therapeutic targets. We used chronic constriction injury (CCI) model mice whether Dex prevents inflammation response. ? 2-adrenoceptors (?2AR) inhibitor BRL44408 adenovirus for knocking down High mobility group box 1 (HMGB1) administrated confirm exert its through targeting ?2AR HMGB1. results indicated that significantly inhibited CCI induced inflammatory response decreasing release mRNA expression IL-1?, IL-6, TNF-? while increasing IL-10. Moreover, participates regulation HMGB1, Toll-like receptor 4 (TLR4), NF?b (p-65) phosphorylation I?B-?. In conclusion, could relieve HMGB1 attenuate
